Application of Auramine O stain: Flood the smear with Auramine O stain and incubate for 15 minutes. Ensure the stain covers the entire smear.Rinse the slide: Rinse the slide with water to remove excess stain.Counterstain: Apply a decolorizing agent such as acid-alcohol to destain the smear. Rinse the slide with water again.
Observation: Examine the slide under a fluorescence microscope. Acid-fast bacteria will fluoresce bright yellow-green when illuminated with an appropriate light source.Always follow the specific protocol provided with the stain and consult appropriate laboratory safety procedures when working with hazardous materials.
